A variety of sterile and non-sterile clinical specimen sources for direct detection of M. tuberculosis complex DNA can be tested. This PCR assay provides a lower limit of sensitivity at 400 cells/mL.

Specify specimen source on the test requisition form and specimen
container. Ship within 24 hrs of collection by overnight courier.
Collect sputum early in the morning from a deep, productive cough
on 3 consecutive days. Place in sterile, leakproof container and
ship ambient or on cold pack.
Other acceptable specimens: Whole blood and bone marrow should be
shipped ambient within 24 hours of collection. Heparinzed blood/bone
marrow is not acceptable.
2.0 (0.5) bronchoalveolar lavage collected in a sterile, leakproof
container. Bronchial wash specimens
are also acceptable if shipped under refrigerated or frozen
2.0 (1.0) pleural, pericardial or gastric fluid or aspirates in
sterile container. If gastric aspirate cannot be shipped immediately,
add 100 mg of sodium bicarbonate as a buffer to neutralize any acid
detrimental to Mycobacteria.
Tissue and needle biopsies: Place in sterile saline.

M. tuberculosis complex consists of the following: M. tuberculosis M. bovis, M. bovis BCG, M. africanum, M. microti, M. canettii. This discrete group of organisms share a DNA homology of greater than 95%. This test is not approved for the testing of patient samples from New York State. This test(s) was performed using a kit that has not been cleared or approved by the FDA. The analytical performance characteristics of this test have been determined by Quest Diagnostics Nichols Institute, Valencia, CA. This test should not be used for diagnosis without confirmation by other medically established means.
